General description

Periostin (POSTN) gene with 23 exons spanning 36 kb is mapped to chromosome 13q13.3. The gene codes for periostin, a matricellular protein that belongs to the family of non-structural matrix proteins. Periostin structure contains a signal peptide, four fasciclin-like domains (FAS1) with vitamin K-dependent γ-carboxyglutamic acid (Gla) residues, and hydrophilic C-terminal region with the heparin-binding site. The protein is mainly expressed in the periosteum, periodontal ligament, heart valves, and tendons. The antibody detects endogenous level of total periostin protein.

Biochem/physiol Actions

Periostin (POSTN) facilitates extracellular matrix structure and organization. It acts as a collagen chaperone and regulates type I collagen fibrillogenesis. POSTN is implicated in early stages of bone formation, tissue repair, and cell recruitment and adhesion. This protein is essential for cancer stem cell maintenance and metastasis. Increased expression of POSTN has been observed in various types of cancers including prostate, breast, lung, pancreatic, ovarian, and colorectal cancer. Hence, the postn levels can be used as a prognostic biomarker for these cancers.

Target description

Binds to heparin. Induces cell attachment and spreading and plays a role in cell adhesion. May play a role in extracellular matrix mineralization.
