Gene Information

General description

KIAA1199 is an endonuclear protein secreted into the extracellular environment. It encodes a 150kDa, inner ear-specific protein consisting of three domains and an N-terminal secretion signal. It is expressed in the inner ear.
Mouse polyclonal antibody raised against a full-length human KIAA1199 protein.

Immunogen

KIAA1199 (AAH20256, 1 a.a. ~ 992 a.a) full-length human protein.

Application

Anti-KIAA1199 antibody produced in mouse is suitable for western blot assay.

Biochem/physiol Actions

KIAA1199 performs in cell signaling, adhesion, migration and proliferation in human cancers. In colon cancer, it is highly expressed in several cells including cytoplasm, perinuclear space and the cell membrane of adenocarcinomas and cochlea. It mediates hyaluronan (HA) depolymerization in an acidic cellular microenvironment such as clathrin-coated vesicles or early endosomes. In addition, it is also associated with the protein binding, transport, and folding; and Ca2+, G-protein, ephrin, and Wnt signaling. It has been reported that KIAA1199 may be negative regulator of the Wnt/CTNNB1 signaling pathway.

Hypoxic stress drives cancer progression by causing a transcriptional reprogramming. Recently, KIAA1199 was discovered to be a cell-migration inducing protein (renamed CEMIP) that is upregulated in human cancers. However, the mechanism of induction of CEMIP in cancer was hitherto unknown.

Recently, we disclosed that KIAA1199-mediated hyaluronan (HA) depolymerization requires an acidic cellular microenvironment (e.g. clathrin-coated vesicles or early endosomes), but no information about the structural basis underlying the cellular targeting and functional modification of KIAA1199 was available. Here, we show
