General description

PCAF (p300/CBP associated factor, P/CAF), a type A histone acetyltransferases (HAT), has been identified in several species. Human PCAF is a ubiquitous protein member of the GCN5-related N-acetyltransferase (GNAT) superfamily.

Specificity

Anti-PCAF recognizes human and dog PCAF. In some extract preparations, additional band(s) may appear. Staining of PCAF band(s) by immunoblotting is inhibited by the PCAF immunizing peptide.

Immunogen

This sequence is identical to mouse PCAF and varies by 2 amino acids residues in chicken.
synthetic peptide corresponding to the C-terminal (amino acids 817-832) of human PCAF with N-terminal added cysteine, conjugated to KLH.

Application

Anti-PCAF antibody is also suitable for use microarray, immunoblot, chemiluminescent western blot (1:500 using whole extracts of dog MDCK kidney cells), and immunoprecipitation (5-10 μg using RIPA lysate with approx. 4x10,000,000 human acute T cell leukemia Jurkat cells).

Biochem/physiol Actions

PACF (p300/CBP association factor) functions as a transcriptional coactivator in association with p300 and CBP (CREB-binding protein). PCAF share high degree of sequence similarity with human and mouse GCN5 HAT (Histone acetyltransferases). PACF mediates the acetylation of core histone. It is also known for the acetylation of non-histone proteins such as p53, MyoD, TFIIE-b, TFIIE, GATA-1, EKLF, HMGI (Y), and several other cellular and viral proteins. PCAF possesses chromatin modification activity and modulation of transcription factors for the activation of transcription. It also promotes cell growth and development as myogenesis and also contribute to receptor-mediated cell signaling or activation.

Physical form

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Storage and Stability

For continuous use, store at 2-8 °C for up to one month. For prolonged storage, freeze in working aliquots at -20 °C. Repeated freezing and thawing, or storage in frost-free freezers, is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilutions should be discarded if not used within 12 hours.
