General description

Vacuolar protein sorting-associated protein 72 homolog (UniProt: Q15906; also known as Protein YL-1, Transcription factor-like 1) is encoded by the VPS72 (also known as TCFL1, YL1) gene (Gene ID: 6944) in human. YL-1 is a nuclear protein that may serve as a DNA-binding transcriptional regulator and may be involved in chromatin modification and remodeling. YL-1 is plays multiple roles in chromatin modification and remodeling in cells and it is shown to be a subunit of TRRAP/TIP60 HAT complex and of chromatin remodeling SCCAP-containing complex. It has a DNA binding region that includes amino acids 156 to 206. In addition, it has an aspartic/glutamic acid rich acidic region (aa 22-77) and a proline rich region (aa 333-354). Two isoforms of YL-1 have been described that are produced by alternative splicing. (Ref.: Cai, Y., et al. (2006). J. Biol. Chem. 280 (14); 13665-70).

Clone EX-1Y32 detects Vacuolar protein sorting-associated protein 72 homolog (YL-1) in human and murine cells.

GST-tagged full-length recombinant human YL-1 protein.

Western Blotting Analysis: A representative lot detected YL-1 in Western Blotting applications (Obri, A., et. al. (2014). Nature. 505(7485):648-53).

Immunocytochemistry Analysis: A 1:800 dilution from a representative lot detected YL-1 in HeLa cells.

Western Blotting Analysis: 2 µg/mL from a representative lot detected YL-1 in 10 µg of HeLa cell lysate.

Immunoprecipitation Analysis: 1-10 µg from a representative lot detected YL-1 in Mouse embryonic fibroblast (MEF) total cell extracts.

Quality

Evaluated by Western Blotting in Mouse embryonic fibroblast (MEF) cell lysate.

Western Blotting Analysis: 1 µg/mL of this antibody detected YL-1 in Mouse embryonic fibroblast (MEF) cell lysate.

Target description

~48 kDa observed, 40.59 kDa calculated. Uncharacterized bands may be observed in some lysate(s).

Storage and Stability

Stable for 1 year at -20°C from date of receipt. Handling Recommendations: Upon receipt and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance.
