The Amicon Ultra products are designed to retain, not filter, and allow high recovery of smaller molecules. For example, let's say you have a 120 kDa molecule and you use a 50 kDa mwco filter in the Amicon Ultra. Most large molecules will be recovered, even 90-98%. However, depending on the size of a small molecule, it is hard to determine if 20-80% of something smaller will pass through. Shape matters and size matters. In this example, if you had a 1 kDa small molecule and a 20 kDa, one may expect more of the 1 kDa to pass through and less of the 20 kDa, if any. One key can sometimes be to spin slower and longer. By doing this, more of the small may pass through.

General description
The vertical design and available membrane surface area provide fast sample processing, high sample recovery (typically greater than 90% of dilute starting solution), and the capability for 80-fold concentration. Solute polarization and subsequent fouling of the membrane are minimized by the vertical design, and a physical deadstop in the filter device prevents spinning to dryness and potential sample loss.
The concentrate is collected from the filter device sample reservoir using a pipetter, while the ultrafiltrate is collected in the provided centrifuge tube. The device can be spun in a swinging bucket or fixed angle rotor. With their vertical membrane design and deadstop, Amicon®Ultra 15 mL filters deliver the highest recovery with short spin times.

Can product UFC505008, Amicon® Ultra Centrifugal Filter, 50 kDa MWCO effectively concentrate the AAV virus sample ? Is it recommended to block the membrane before adding the sample as AAV strongly adhere to the column?

Yes, product UFC505008 ,Amicon Ultra Centrifugal filter is suitable for concentrating the AAV virus up to a certain point. However, to optimize the process and prevent aggregate formation, it is recommended to use larger volume Amicon Ultras (15 ml) and avoid over-concentration.

My antibody solution contains 20% glycerol. Can I use this poduct to get rid of the glycerol? which dialysis buffer should be used?

Yes, Amicon products that have a 10 kDa NMWCO pore size are suitable for the removal of glycerol from antibody preparations. The protocol below was provided by one of our antibody suppliers - Atlas Antibodies. Note that this protocol is written for the 0.5 mL product UFC501096, Volumes and possibly spin times would have to be adjusted to accommodate the 15 mL Amicon products.
Glycerol can be removed using Amicon Centrifugal Filter Units, 10 kDa cutoff (Merck Millipore UFC501096).
The filter unit concentrates the antibody sample during centrifugation by allowing the buffer to flow through the membrane but stops any proteins above 10 kDa from passing.The antibody is then resuspended in a buffer of choice, thus allowing for buffer exchange. Consider testing the antibody +/- the glycerol cleanup, to see whether there is an improvement. Some antibodies perform better even if there is glycerol in the buffer.
PROCEDURE
1. Dilute the antibody to 400 μl with sterile PBS (or other buffer of choice).
2. Place the Amicon filter unit into the supplied collection tube.
3. Add 400 μl of diluted antibody to the filter unit. Close the lid.
4. Spin for 5-10 minutes at 14k*g or until 100 μl of the antibody sample remains in the spin column. If this has not been achieved during the first 5 min, then centrifuge for a few more minutes until only 100 μl remains.
Please note that the suggested time it takes to concentrate the sample down to 100 μl should be seen only as a guideline and will ultimately have to be determined by the user. For all spins, place the filter part of the filter unit and the elbow of the collection tube towards the centre of the rotor. All spins are performed at room temperature.
5. Discard the flow through.
6. Add 300 μl of sterile PBS (or other buffer of choice) to the filter unit and centrifuge again as outlined in step 4. Then discard the flow through.
7. Repeat step 6 four more times until the glycerol content is 0,02 %. 5-minute centrifugations are usually enough to reach 100 μl left in the filter unit at these steps. In the last round, centrifuge for 4 minutes until 100 μl is reached.
8. In new collection tube, place spin column upside down.
9. Spin for 2 min at 1k*g to collect buffer-exchanged antibody.Helpful?

Unfortunately, this is the only 15mL centrifugal filter with 50kDa MWCO offered. There are other products available at different MWCO or volumes such as the Corning Spin-X UF products, see CLS431490 (20 mL).
