For the cytokine analysis of brain tissue using Milliplex Cytokine kits , the following protocol is recommended:
Tissue Collection:
•Extract the brains from each animal during euthanasia.
•Cut the tissue into small pieces using two scalpel blades.
•Shock-freeze approximately 0.2 ml of tissue in liquid nitrogen or using dry ice.
•Store the frozen tissue at -80°C until further processing for cytokine analysis.
Tissue Homogenization:
•Homogenize the tissue in 1 ml of PBS containing protease inhibitors (the ratio of tissue to volume should be 10 to 1, for example, 1 ml for 100 mg of tissue).
•Centrifuge the homogenates at a speed greater than 10,000g for at least 10 minutes.
•Use the supernatants for cytokine assays or store them at -80°C.
Dilutions of the homogenates should be determined as necessary.

General description
Cytokines are immunomodulatory polypeptides that play key roles in both adaptive and innate immune responses.“Cytokine” is a general term used for a diverse group of soluble proteins and peptides which act as regulators under both normal and pathological conditions to modulate the functional activities of individual cells and tissues. These proteins also mediate direct interactions between cells and regulate processes taking place in the extracellular environment. Application
