General description

MUC3A (mucin 3A) belongs to the family of epithelial mucins, which are both secreted as well as cell surface glycoproteins. This family is expressed by epithelial tissues, and MUC3A is predominantly expressed in the small intestine. This gene maps to human chromosome 7q22. MUC3A contains an extracellular region with two cysteine-rich EGF (epidermal growth factor)-like domains, intervened by an N-glycosylation-rich domain, along with a putative coiled-coil domain. This is succeeded by a transmembrane domain, and a cytoplasmic region consisting of 75 amino acids. MUC3A has an alternatively spliced isoform which contains the first EGF- like domain, but lacks the second EGF-like domain and the transmembrane region.

Application

Biochem/physiol Actions

Mucin family of proteins is involved in epithelial cell protection, regulation of adhesion and cell signaling. MUC3A (mucin 3A) confers protection to intestinal epithelial cells against the external environment. Variations in this gene determine the susceptibility of an individual to ulcerative colitis and Crohn′s disease (CD). This protein is expressed in synovial lining cells, macrophages, and fibroblasts, and it is overexpressed in synovial tissue. Therefore, it might play a role in the pathogenesis of arthritis. Hypomethylation of the 5′-flanking region of MUC3A gene determines the expression of this gene in various cancers such as breast, pancreas, colon and lung cancer cell lines. Therefore, methylation studies of this gene might help in predicting cancer risk as well prognosis of cancer patients. This gene is also up-regulated in renal clear cell carcinoma, and is involved in tumorigenesis of renal cancer.
