General description

A metachromatic dye suitable for a wide variety of histological staining procedures.

Application

Toluidine Blue O along with light-emitting diode has been used as a photosensitizer to control in vitro biofilms and to kill gram-negative bacteria.[1][2]

Biochem/physiol Actions

Toluidine Blue O (TBO) is a thiazine dye of the quinone-imine family and is cationic in nature. It has been employed in polychromatic staining of paraffin embedded plant cell walls.[3] TBO can be removed from its aqueous solution by using an effective adsorbent, clinoptilolite.[4] TBO and fixed output laboratory laser light source are used together to destroy variety of oral bacteria associated with dental caries.[5] This metachromatic nuclear stain is often used in place of thionin or methylene blue. It also serves as a component of a tribasic stain along with Malachite Greenand Basic Fuchsin.[6]

Suitability

Certified for use as a metachromatic stain for mast cells and cartilage using the Churukian-Schenk method and the method of Nocht on paraffin sections.

Questions

1–2 of 2 Questions
  1. Is there a recommendation for dissolving Toluidine Blue (198161-5g lot MKCK0257) at 1 mg/ml in water?

    1 answer
    1. This product is released based on the testing performed by the Biological Stain Commission. There is no specific solubility specification for this product. At the BSC, the product is dissolved at 50 mg in 250 ml of water for one test, and 100 mg of the dye is dissolved in 150 ml of water for the second test, followed by the addition of additional alcohol. When used to stain frozen sections, a 0.5% solution is typically prepared in 20% alcohol. Some procedures call for dissolving at 10 mg/ml, but this amount of dye cannot always be dissolved. Once dissolved, the solution should be relatively stable. This product does not appear to be hygroscopic, and its solubility can vary somewhat from lot to lot.

  2. Could you provide the protocol for using Toluidine Blue for MMA sections?

    1 answer
    1. There are no  protocols for using Toluidine Blue O to stain MMA sections.
