Specificity

Clone 1C7-H1 ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Pyruvate dehydrogenase kinase 1 (PDH kinase 1). It targets an epitope within 15 amino acids from the C-terminal region.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the C-terminal region of human Pyruvate dehydrogenase kinase 1 (PDH kinase 1).

Application

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected PDH kinase 1 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected PDH kinase 1 in lysates from RAW264.7 and C6 cells.

Affinity Binding Assay: A representative lot of this antibody bound PDH kinase 1 peptide with a KD of 1.0 x 10-12 in an affinity binding assay.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected PDH kinase 1 in HeLa cells.

Flow Cytometry Analysis: 1 µg from a representative lot detected PDH kinase 1 in one million Hela cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected PDH kinase 1 in Human cerebral cortex tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Pyruvate dehydrogenase (acetyl-transferring)] kinase isozyme 1, mitochondrial (UniProt: Q15118; also known as EC:2.7.11.2, Pyruvate dehydrogenase kinase isoform 1, PDH kinase 1) is encoded by the PDK1 (also known as PDHK1) gene (Gene ID: 5163) in human. PDH kinase 1 is a mitochondrial matrix enzyme that is synthesized with a transit peptide (aa 1-28), which is subsequently cleaved off. It is expressed predominantly in the heart and is detected at lower levels in liver, skeletal muscle, and pancreas. It plays a key role in regulation of glucose and fatty acid metabolism and homeostasis via phosphorylation of the pyruvate dehydrogenase subunits PDHA1 and PDHA2. This inhibits pyruvate dehydrogenase activity, and thereby regulates metabolite flux through the TCA cycle, down-regulates aerobic respiration and inhibits the formation of acetyl-CoA from pyruvate. It is also reported to play an important role in cellular responses to hypoxia and is shown to be important for cell proliferation under hypoxic conditions. It also protects cells against apoptosis in response to hypoxia and oxidative stress. Its levels are up-regulated via the HIF1 signaling in response to hypoxia. Exposure of cancer cells to severe hypoxia is reported to induce translocation of AKT to the mitochondrion, which leads to AKT-mediated phosphorylation of PDH kinase 1 at threonine 346 that supports tumor cell survival and proliferation during hypoxia. PDH kinase 1 is phosphorylated by constitutively activated Abl1, FGFR1, FLT3 and JAK2. Its phosphorylation at tyrosine 243 and 244 strongly increases its kinase activity, while phosphorylation at tyrosine 136 has reduced effect. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonal. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
