General description

α-Glucosidase hydrolyzes carbohydrates by acting on terminal, non-reducing a(1→4)-linked D-glucose residues with the release of D-glucose.[1] Defects in α-glucosidase have been implicated in Pompe disease and diabetes.[2]

Principle

In this assay,α-glucosidase activity is determined by a reaction in which α-glucosidase hydrolyzes p-nitrophenyl-α-D-glucopyranoside resulting in the formation of a colorimetric (405 nm) product, proportional to the α-glucosidase activity present

  1. about cat # MAK123, in the the protocol, the samples are incubated either at room temperature or 37°C and then absorbance is measured after 20 min. Please indicate me the incubation time at RT and incubation time at 37C because kinetic is different at T

    1. The calibrator is incubated under the same conditions as the sample, and the activity is calculated relative to the absorbance of the calibrator, so it doesn't matter what temperature is used for incubation.

  2. Hi, I have a question about cat # MAK123, a-glucosidase activity assay kit, which is sufficient for 100 assays. Once the reagents are opened, can they be used at a later date? eg - am I able to open the kit and run samples over several days?

    1. The component included in this kit may be refrozen for later use until the expiration date. Do not store refrigerated.
