General description

Up-frameshift suppressor 3 homolog A (UPF3A) shuttles between nucleus and cytoplasm. In humans there are two paralogs of UPF3, UPF3A and UPF3B. UPF3A transcript is highly expressed in testis, uterus, muscle, fetal brain and spinal cord.

Application

Biochem/physiol Actions

Up-frameshift suppressor-3 homolog A (UPF3A) is involved in nonsense-mediated decay (NMD), an RNA decay pathway. A mutant of UPF3A which is unable to exit the nucleus does not participate in nonsense-mediated RNA decay pathway. UPF3A levels are tightly regulated in cells by its destabilization via UPF3B-dependent mechanism. UPF3A forms a complex with RNA-binding protein 8A (Y14), protein mago nashi homolog (MAGOH), eukaryotic initiation factor 4A-III (eIF4AIII) and barentsz (BTZ). Together the complex is required for NMD pathway. UPF3A interacts specifically with spliced mRNAs. It is also a component of exon-exon junction complex and is important for translation efficiency.
