Cell Line Description
HR5-CL11 cells were obtained after transfection of HeLa cells with plasmids coding for a transcriptional transactivator and transferring G418 and hygromycin resistance. This tetracycline-controlled transactivator (a fusion between the tet repressor and the activating domain of the herpes simplex virus VP16 transcription activation domain) can stimulate the expression of a reporter gene up to 100,000 fold. Subsequently these cells were stably transfected with a luciferase reporter unit. Luciferase expression in the HR5-CL11 cells is highly stimulated by doxycycline.

Culture Medium
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Subculture Routine
Split sub-confluent cultures (70-80%) 1:5 to 1:20 i.e. seeding at 2x1,000 to 2x10,000 cells/cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C.
