General description

Amicon® Ultra 15 mL centrifugal devices with Ultracel® 30,000 Dalton molecular weight cutoff filters provide fast ultrafiltration, and reliable retention of macromolecules. For example, Ovalbumin (45 kDa) at 1 mg/ml initial concentration is retained at >95% .

The vertical design and available membrane surface area provide fast sample processing, high sample recovery (typically greater than 90% of dilute starting solution), and the capability for 80-fold concentration. Solute polarization and subsequent fouling of the membrane are minimized by the vertical design, and a physical deadstop in the filter device prevents spinning to dryness and potential sample loss.

The concentrate is collected from the filter device sample reservoir using a pipetter, while the ultrafiltrate is collected in the provided centrifuge tube. The device can be spun in a swinging bucket or fixed angle rotor. With their vertical membrane design and deadstop, Amicon® Ultra 15 mL filters deliver the highest recovery with short spin times.

Application

  • Ultrafiltration with 30,000 Dalton molecular weight cutoff Ultracel® membrane.
  • Concentration of biological samples containing antigens, antibodies, enzymes, nucleic acids, or microorganisms.
  • Purification of macromolecular components found in tissue culture extracts or cell lysates and protein removal prior to HPLS.
  • Concentration of dilute or pre-purified proteins from column eluents.
  • Desalting, buffer exchange and protein dialysis.

  1. How many times can sample be added to the same filter unit? For example: If I have 100 mL cell free supernatant and want to add 12 mL at a time, centrifuge and collect retentate - followed by discarding the flow through and pouring the next 12 mL etc.

    1. The same Amicon filter unit can be used for repeated spins with the same sample. However, repeated spins may lead to increased clogging, resulting in slower flow rates and longer spin times.

  2. I noticed no volume change after centrifuging 10mL cell media at 14,000xg for 20minutes. Could this be due to unsuccessful concentration?

    1. The lack of volume change suggests that the sample may contain a high amount of particulate matter, leading to rapid clogging and preventing effective concentration.

  3. Hello! Our lab had to move to a different area of our building and we found some probably quite old boxes of Amicon Ultra 15s (Cat UFC903096, lot R8CN67048). Does this product have an expiration date/shelf life? If so, has this lot surpassed it?

    1. This product is not assigned an expiration date or recommended retest date. Products with no expiration date or recommended retest date should be routinely inspected by customers to ensure they perform as expected. These products are also subject to a one-year warranty from the date of shipment.
