General description

Titin is a large complex protein containing over one hundred IgG-like and fibronectin type-III domains, in addition to multiple Kelch, RCC1, TPR, and WD repeats. It is a member of the CAMK Ser/Thr protein kinase family and also possesses a protein kinase domain, which is involved in controlling gene expression and protein turnover in muscle tissues. Titin also functions as a sensor to mediate appropriate cellular responses to mechanical stress. In striated muscle tissue, titin regulates the length of the sarcomere by integrating myosin and actin filaments at the Z-disk and M-band, and facilitates the elasticity of the sarcomere. Titin also regulates sarcomere assembly and functions as central hub in signal transduction pathways. Defects in Titin may contribute to various forms of cardiomyopathy.
