General description

Probable ATP-dependent RNA helicase DDX41 (UniProt: Q9UJV9; also known as DEAD box protein 41; DEAD box protein abstrakt homolog) is encoded by DDX41 (also known as ABS) gene (Gene ID 51428) in human. DDX41 belongs to the family of DEAD box helicase family of proteins, characterized by the presence of the conserved Asp-Glu-Ala-Asp (DEAD) motif. DDX41 is reported to sense DNA virus in myeloid dendritic cells (mDCs) mediated by the adaptor STING (stimulator of interferon genes). DDX41 is reported to bind to both DNA and STING through a DEADc domain and then DDX41-STING complex localizes to the cytoplasm. DDX41 binds bacterial DNA in the infected cells and is shown to be more important than gamma-interferon-inducible protein IFI16 in the initial sensing of cytosolic B-form DNA in triggering the early burst of type I interferon response. Knockdown of DDX41 expression blocks the ability of mDCs to mount type I interferon and cytokine responses to DNA and DNA viruses. Ref.: Zhang, Z., et al. (2011). Nat. Immunol. 12(10): 959-966.

Specificity

Clone 4D8.2 detects DDX41 protein in HEPG2 cells by Western blotting and in human ovarian and cervical cancer tissue by immunohisotchemistry.

Immunogen

GST-tagged recombinant fragment corresponding to 115 amino acids from the N-terminal region of human DDX41.

This mouse monoclonal Anti-DDX41 antibody, clone 4D8.2, Cat. No. MABF1107 is validated for use in Immunohistochemistry (Paraffin) and Western Blotting for the detection of DDX41.
Western Blotting Analysis: 0.5 µg/mL from a representative lot detected DDX41 in 10 µg of HeLa cell lysate.

Immunohistochemistry Analysis: A 1:50 dilution from a representative lot detected DDX41 in human ovarian cancer, human cervical cancer, and human prostate cancer tissue.




Quality

Evaluated by Western Blotting in HEPG2 cell lysate.

Western Blotting Analysis: 0.5 µg/mL of this antibody detected DDX41 in 10 µg of HEPG2 cell lysate
