General description

Thrombopoietin (TPO) also known as c-Mpl ligand is an acidic glycoprotein expressed mainly in the liver, kidney, and bone marrow. Recombinant rat TPO is a fully biologically active 174 amino acid polypeptide (18.7kDa), which contains the erythropoietin-like domain of the full length TPO protein.

Application

Thrombopoietin (TPO) from rat has been used in experimental systems to determine the effect of TPO in recovery from thrombocytopenia.

Biochem/physiol Actions

Thrombopoietin (TPO) acts as a key physiologic regulator of megakaryocyte and platelet production in vivo. Mutation of TPO or its receptor might lead to thrombocytopenia and megakaryocytopoiesis. In addition, TPO also influence the action of erythroid and erythromegakaryocytic progenitors in mouse embryonic yolk sac. TPO binds to c-mpl receptor and regulates circulating platelet level.

Physical form

Lyophilized from 5 mM Sodium Phosphate, pH 7.6 + 75 mM NaCl.

Reconstitution

Centrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/ml. Do not vortex. This solution can be stored at 2-8°C for up to 1 week. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C.
