Especificidad

Clone 3I17 is a ZooMAb® recombinant rabbit monoclonal antibody that specifically detects Ribosomal Protein S3. It targets an epitope within 19 amino acids from the C-terminal region.

Aplicación

Quality Control Testing

Evaluated by Western Blotting in HeLa cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Ribosomal Protein S3 in HeLa cell lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected RPS3 in lysates from HEK-293 and PC-12 cells.

Affinity Binding Assay: A representative lot of this antibody bound RPS3 peptide with at least one thousand-fold (1,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected RPS3 in HeLa cells.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected RPS3 in human kidney tissue sections.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Small ribosomal subunit protein uS3 (UniProt: P23396; also known as 40S ribosomal protein S3) is encoded by the RPS3 gene (Gene ID: 6188) in human. RPS3 (ribosomal protein S3) codes for a translational protein which belongs to the ribosomal small subunit protein. It is localized to the surface of 40s ribosomal subunit. The RPS3 gene is mapped to human chromosome 11q13.4. This gene is co-transcribed with the small nucleolar RNA genes U15A and U15B, which are located in its first and fifth introns, respectively. As is typical for genes encoding ribosomal proteins, there are multiple processed pseudogenes of this gene dispersed through the genome. The ribosomal protein S3 belongs to the S3P family of ribosomal proteins and appears to be located in both the cytoplasm and can translocate to the nucleus, particularly in response to DNA damage, where it participates in DNA repair processes. Alternatively spliced transcript variants encoding 2 different isoforms have been found for this gene. RPS3 is ubiquitously expressed across various tissues with higher expression levels typically observed such as the liver, pancreas, and testis. RPS3 is a multifaceted protein with crucial roles in ribosomal function, DNA repair, apoptosis, and transcription regulation. It has been demonstrated that the protein has an extraribosomal role as an endonuclease involved in the repair of UV-induced DNA damage. Higher levels of expression of this gene in colon adenocarcinomas and adenomatous polyps compared to adjacent normal colonic mucosa have been observed. Studies suggest that Lnc-SNHG5 promoted Hepatocellular Carcinoma progression through the RPS3-NF B Pathway. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Forma física

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitución

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Almacenamiento y estabilidad

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
