ZooMAb antibodies represent an entirely new generation of recombinant monoclonal antibodies.

Each ZooMAb antibody is manufactured using our proprietary recombinant expression system, purified to homogeneity, and precisely dispensed to produce robust and highly reproducible lot-to-lot consistency. Only top-performing clones are released for use by researchers. Each antibody is validated for high specificity and affinity across multiple applications, including its most commonly used application. ZooMAb antibodies are reliably available and ready to ship when you need them.

Especificidad

Clone 5D17 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Rhodopsin. It targets an epitope with in 16 amino acids from the N-terminal, extracellular domain.

Inmunógeno

KLH-conjugated linear peptide corresponding to 16 amino acids from the N-terminal region of human Rhodopsin.

Aplicación

Quality Control Testing

Evaluated by Western Blotting in Mouse Eye tissue lysate.

Western Blotting Analysis: A 1:1,000 dilution of this antibody detected Rhodopsin in Mouse Eye tissue lysate.

Tested Applications

Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected Rhodopsin in human retina tissue lysate.

Affinity Binding Assay: A representative lot of this antibody bound Rhodopsin peptide with a KD of 1.5 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Rhodopsin in human retina tissue sections.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Rhodopsin in Y79 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user
Anti-Rhodopsin, clone 5D17 ZooMAb®, Cat. No. ZRB1157, is a recombinant Rabbit monoclonal antibody that specifically targets Rhodopsin and is tested in Affinity Binding Assay, Immunocytochemistry, Immunohistochemistry (Paraffin) and, Western Blotting.

Descripción de destino

Rhodopsin (UniProt: P08100; also known as Opsin-2) is encoded by the RHO (also known as OPN2) gene (Gene ID: 6010) in human. Rhodopsin is a multi-pass membrane protein that is found in rod shaped photoreceptor cells that mediate vision in dim light. It contains four extracellular domains, seven transmembrane domains, and four cytoplasmic domains. It is synthesized in the inner segment of rod photoreceptor cells before vectorial transport to disk membranes in the rod outer segment photosensory cilia. Rhodopsin is required for image-forming vision at low light intensity. It is required for photoreceptor cell viability after birth. Light-induced isomerization of the chromophore 11-cis-retinal to all-trans-retinal triggers a conformational change that activates signaling via G-proteins. Subsequent receptor phosphorylation mediates displacement of the bound G-protein alpha subunit by the arrestin SAG and terminates signaling. Mutations in RHO gene are known to cause Retinitis pigmentosa 4 that is characterized by retinal pigment deposits and loss of rod photoreceptor cells followed by secondary loss of cone photoreceptors leading to night vision blindness. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Forma física

Purified recombinant mouse monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.

Reconstitución

30 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Almacenamiento y estabilidad

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
