General description

The F2 gene encodes thrombin protein and is mapped to human chromosome 11p11.2.

Application

Thrombin from human plasma has been:
  • dissolved in CaCl2 and added to poly ethylene glycolated fibrinogen/cell solution for fibrin production
  • to polymerize fibrin on well plates
  • used as an endogenous agonist proteinase for proteinase-activated receptors : PAR1, PAR3 and PAR4 for stimulation of cells

Biochem/physiol Actions

Thrombin is an endolytic serine protease which stimulates vascular damage induced-leukocytes, platelets, endothelial and mesenchymal cell production. This action is mediated via G-protein-coupled receptor. It catalysis the cleavage of fibrinogen at Arg−Gly bonds to form fibrin and release fibrinopeptides A and B. The predominant form of thrombin in vivo is the zymogen prothrombin (factor II), which is produced in the liver. Prothrombin is a coagulation activation factor, high levels of which indicates cancer. Thrombin can be used to cleave fusion proteins. Cleavage of fusion proteins can be carried out at a thrombin:fusion protein ratio of 1:500.

Lyophilized from saline sodium citrate buffer, pH 6.5

Analysis Note

The NIH assay procedure uses 0.2 ml diluted plasma (1:1 with saline) as a substrate and 0.1ml of thrombin sample (stabilized in a 1% buffered albumin solution) based on a modification of the method of Biggs. Only clotting times in the range of 15-25 seconds are used for determining thrombin concentrations.

    1. Prior to lyophilization the thrombin is at concentration of 1250-1450 units/mL in a solution of saline sodium citrate buffer, pH 6.5.

  2. Hello! How long can we store the lyophilised powder in stock solutions at -20°C?

    1. Stability of thrombin in solution is not evaluated. However, external reports have shown a 10% loss of activity per year when reconstituted at a concentration of 1mg/mL and stored at -20C. Freeze/thaw cycles should be avoided.

  3. Which volume of liquid do I have to add to a 100 U T7009 vial? And T6884 vial? With which kind of liquid should the powder be dissolved?

    1. The product is soluble in water (10 mg/mL). Stock solutions can also be prepared at a concentration of 100 units/mL in a 0.1% (w/v) BSA solution. Stock solutions remain active for one week at 0–5 °C. Solutions are most stable at pH 6.5, as a pH >7 will greatly reduce thrombin activity. Since thrombin solutions adsorb to glass, it is recommended to aliquot the solutions in plastic tubes and store at –20 °C for long-term storage. We can calculate the weight in MG for this lot of 250 UN product using lot specific information found on a CoA (Refer to a certificate).

      For T7009-250UN for example: As per one lot in one CoA, units/mg is 1742.
      1742 units = 1 mg
      250 units = 1mg/1742 x 250 = 0.1435 mg
      So, 250 units of this product lot contains 0.1435 mg (143.5 ug) of powder in vial.

    1. Both of these Thrombin products - T7009 and T6884 - are sold by activity or units. The amount of powder in the vial is lot specific and based on the activity reported in the Certificate of Analysis.

      T6884-100UN, lot SLCP0517 tested at 2736 uits per milligram. The 100UN vial contains 36ug of powder per vial. See the link below to review this Certificate of Analysis:

    1. Thrombin from human plasma gave a clear very faint yellow solution when dissolved at 100 unit vial plus 1 ml of 28 mM sodium barbital, 0.125M sodium chloride, 1% BSA, 0.5% PEG, pH 7.35.  Thrombin from bovine plasma (T4648) is soluble in water at 10 mg/mL.

Thrombin from any vertebrate species will clot the fibrinogen of virtually any other species.  Fibrinogen from any mammalian source will cross-react with thrombin from any mammalian source, but it is not well known how thrombin and fibrinogen from different animals differ in their sequence.  When any mammalian thrombin is injected into a different mammal, clotting will occur.
