General description

This product is extracted from E. coli serotype O55:B5 and purified by gel filtration. The source strain is CDC 1644-70. The LPS O55:B5 has been used to stimulate human peritoneal macrophages at 1 ng/ml and to stimulate equine peritoneal macrophages at 1-100 ng/ml.

Application

Lipopolysaccharides (LPSs) are characteristic components of the cell wall of Gram-negative bacteria. LPS and its lipid A moiety stimulate cells of the innate immune system by the Toll-like receptor 4 (TLR4), a member of the Toll-like receptor protein family, which recognizes common pathogen-associated molecular-patterns (PAMPs).

Biochem/physiol Actions

Lipopolysaccharides (LPS) are localized in the outer layer of the membrane and are, in noncapsulated strains, exposed on the cell surface. They contribute to the integrity of the outer membrane, and protect the cell against the action of bile salts and lipophilic antibiotics.

Preparation Note

The product is soluble in water (5 mg/ml) or cell culture medium (1 mg/ml) yielding a hazy, faint yellow solution. A more concentrated, though still hazy, solution (20 mg/ml) has been achieved in aqueous saline after vortexing and warming to 70-80 oC. Lipopolysaccharides are molecules that form micelles in every solvent. Hazy solutions are observed in water and phosphate buffered saline. Organic solvents do not give clearer solutions. Methanol yields a turbid suspension with floaters, while water yields a homogeneously hazy solution.

Preguntas

1–5 de 5 Preguntas
  1. Wie lässt sich eine wässrige Lösung steril filtrieren? Welches Filtermaterial, welche Porengröße?

    1 respuesta
    1. An aqueous solution can be sterile filtered using a filter with a pore size of 0.2 µm. A low protein binding membrane, such as polyvinylidene fluoride (PVDF) or polyethersulfone (PES), is recommended for this process.

  2. How can I determine the shelf life / expiration / retest date of this product?

    1 respuesta
    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.
For products without retest or expiration dates, our standard warranty of 1 year from the date of shipment is applicable.

  3. How is shipping temperature determined? And how is it related to the product storage temperature?

    1 respuesta
    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers. For more information, please refer to the Storage and Transport Conditions document: https://www.sigmaaldrich.com/deepweb/assets/sigmaaldrich/marketing/global/documents/316/622/storage-transport-conditions-mk.pdf

  4. What is the molecular weight of this product ?

    1 respuesta
    1. Lipopolysaccharides(LPS) are heterogeneous and do not have fully defined molecular weights. In the presence of strong surface active agents, and in the absence of divalent cations, LPS will have molecular weights in the range of 10?20 kDa. Please see the link below to review additional information regarding the nature of LPS, including molecular weight variability:

  5. Can I use DPBS to dilute? or just use water?

    1 respuesta
    1. This product is tested for solubility at 5 mg/mL in water. Aqueous buffers such as DPBS are expected to be suitable as well. Pease see the link below to review a reference that may be helpful:
