General description
Siembra en placa de células nutritivas MEF
Procedimiento:
1. Antes de descongelar las células nutritivas PMEF, cubra las placas/matraces con una disolución de gelatina.
2. Descongele rápidamente los viales de PMEF en un baño María a 37 °C y transfiéralo a un tubo de 15 ml (que ya contiene 10 ml de medio nutritivo celular PMEF caliente). Invierta suavemente el tubo para distribuirlo y centrifugue a 300 x g durante 4–5 minutos.
3. Retire el sobrenadante y vuelva a suspender el sedimento celular en el medio nutritivo celular PMEF caliente.
4. Retire la disolución de gelatina de las placas/matraces y distribuya en alícuotas la suspensión nutritiva celular PMEF a las densidades recomendadas en la Tabla 4.1 de la guía de protocolo ES de ratón
5. Incube las células nutritivas PMEF a 37 °C con CO2 al 5 %. Utilice las figuras 4A, B y C del protocolo ES de ratón como guía para calcular la densidad y
el aspecto correctos de PMEF. Las placas gelatinizadas se pueden utilizar durante 12–14 días.
