General description
SRC-1 is a member of a gene family that functions in the full transcriptional activation of the steroid hormone receptor superfamily and contains intrinsic histone acetyltransferase activity. SRC-1 has a molecular weight of 160 kDa and has a basic helix-loop-helix (bHLH)-PAS domain.

Immunogen
KLH conjugated synthetic peptide (CKVADFGLARLIEDNEYTARQGA) of Src (avian sarcoma virus), residues 400-422. The immunogen sequence shares 22/23 amino acids with Yes and Fyn, and 21/23 amino acids with Lyn and Hck.
Application
Western Blot Analysis: A 1:2000 to 1:8000 dilution of this lot detected Src in RIPA whole cell lysates from human A431 cells. Immunoprecipitation: 5-20 μL of a previous lot immunoprecipitated Src from 500 μg of A431 RIPA lysate.
Biochem/physiol Actions
Predicted to cross react with frog and chicken. The immunogen sequence shares 22/23 amino acids with Yes and Fyn, and 21/23 amino acids with Lyn and Hck.

Add 2.5 μL of 2-mercaptoethanol/100 μL of lysate and boil for 5 minutes to reduce the preparation. Load 20 μg of reduced lysate per lane for minigels.
