General description

Specificity

Clone 1C4 is a ZooMAb® recombinant rabbit monoclonal antibody that detects Annexin A2. It targets an epitope within 24 amino acids from the internal region.

Immunogen

KLH-conjugated linear peptide corresponding to 24 amino acids from the internal region of human Annexin A2.

Application

Quality Control Testing

Evaluated by Western Blotting in L6 cell lysate.

Western Blotting Analysis (WB): A 1:10,000 dilution of this antibody detected Annexin A2 protein in L6 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Annexin A2 in human and rat lung tissue lysates, and a 1:1,000 dilution detected Annexin A2 in mouse lung tissue lysate.

Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected Annexin A2 in human colon and liver tissue sections.

Immunofluorescence Analysis: A 1:1,000 dilution from a representative lot detected Annexin A2 in human colon tissue sections.

Affinity Binding Assay: A representative lot of this antibody bound Annexin A2 peptide with at least ten thousand-fold (10,000X) higher than with non-specific control peptide.

Immunocytochemistry Analysis: A 1:100 dilution from a representative lot detected Annexin A2 in L6 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

Annexin A2 (UniProt: P07355; also known as Annexin II;Annexin-2;Calpactin I heavy chain;Calpactin-1 heavy chain;Chromobindin-8;Lipocortin II;Placental anticoagulant protein IV (PAP-IV);Protein I;p36) is encoded by the ANXA2 (also known as ANX2, ANX2L4, CAL1H, LPC2D) gene (Gene ID: 302) in human. Annexin A2 belongs to the annexin family of calcium-dependent phospholipid-binding proteins, and is a multifunctional protein involved in a wide range of biological processes. It is characterized by a conserved core domain that mediates membrane binding and a unique N-terminal tail that confers specific protein-protein interactions. The core domain contains four annexin repeats, each forming a five alpha-helix bundle, while the N-terminal tail varies among the annexins and is responsible for the diverse functions of Annexin A2. Annexin A2 functions both intracellularly and extracellularly. Intracellularly, it plays a role in various cellular processes including membrane trafficking, endocytosis, exocytosis, and the organization of the actin cytoskeleton. It acts as an anticoagulant by inhibiting prothrombin activation. Extracellularly, Annexin A2 can act as a receptor for plasminogen and tissue plasminogen activator on the cell surface, facilitating the conversion of plasminogen to plasmin, a key enzyme in fibrinolysis and wound healing. Annexin A2 has been implicated in a variety of diseases. Its role in promoting angiogenesis and stabilizing blood vessels links it to cancer, where it can influence tumor growth, metastasis, and angiogenesis. In cardiovascular disease, its anticoagulant properties are significant, and alterations in its function or expression levels can lead to thrombotic events. Additionally, its involvement in fibrinolysis and inflammation connects it to autoimmune diseases and conditions such as rheumatoid arthritis. Annexin A2 has also been studied in neurodegenerative diseases, where its interaction with cytoskeletal elements suggests a potential role in neuroinflammation and cell death. This ZooMAb® recombinant monoclonal antibody, generated by our proprietary technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals.

Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
