Specificity
Clone 1H14 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Serum response factor (SRF). It targets an epitope within 21 amino acids from the C-terminal half.
Immunogen
KLH-conjugated linear peptide corresponding to 21 amino acids from the C-terminal half of human serum response factor (SRF).
Application
Quality Control Testing
Evaluated by Western Blotting in HeLa cell nuclear extract.
Western Blotting Analysis: A 1:1,000 dilution of this antibody detected serum response factor (SRF) in HeLa cell nuclear extract.
Tested Applications
Western Blotting Analysis: A 1:1,000 dilution from a representative lot detected serum response factor (SRF) in Rat brain tissue lysate.
Immunohistochemistry (Paraffin) Analysis: A 1:1,000 dilution from a representative lot detected serum response factor (SRF) in Human kidney tissue sections.
Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected serum response factor (SRF) in HeLa, HepG2, and NIH3T3 cells.

Target description
Serum response factor (UniProt: P11831; also known as SRF) is encoded by the SRF gene (Gene ID: 6722) in human. SRF is a transcription factor that binds to a serum response element (SRE) associated with a variety of genes, including immediate early genes such as c-Fos, FosB, JunB, Egr-1 and -2; neuronal genes such as nurr1 and nur77. It binds to DNA as a dimer and its DNA binding region is localized in amino acids 133-222. By regulating the expression of several genes, it controls cell growth and differentiation, neuronal transmission, and muscle development. Together with Myocardin Related Transcription Factor A (MRTFA) transcription coactivator, it controls the expression of genes regulating cytoskeleton development, morphogenesis, and cell migration. It is also required for cardiac differentiation and maturation. SRF is shown to be activated by a variety of agents, including serum, lysophosphatidic acid, lipopolysaccharide, 12-O-tetradecanoylphorbol-13-acetate (TPA), and cytokines. Keratinocytes in normal human and mouse skin are reported to express high levels of SRF, but its expression is strongly downregulated in the hyperproliferative epidermis of wounded and psoriatic skin. Loss of SRF is known to disrupt epidermal homeostasis. Mice lacking SRF in cardiac tissue display lethal cardiac defects between E10.5 and E13.5. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS, 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.
Reconstitution
0.3 mg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
