General description

HB-EGF (heparin-binding epidermal growth factor-like growth factor) belongs to the EGF family, and was first isolated as a secreted product from cultured human macrophages. It is produced as a precursor transmembrane protein (pro-HB-EGF), which is composed of EGF-like and heparin-binding domains. This precursor protein can be cleaved to produce a mature and soluble HB-EGF (sHB-EGF) form, and both these forms are active. It is expressed in various cell types including endothelial cells, epithelial cells, hematopoietic cells, and vascular smooth muscle cells.

Biochem/physiol Actions

HB-EGF (heparin-binding epidermal growth factor-like growth factor) functions as a mitogen and a chemotactic factor. Studies in experimental necrotizing enterocolitis (NEC) show that enteral administration of this factor confers protection to intestines from tissue injury, facilitates migration and proliferation of enterocytes, and maintains intestinal stem cell (ISC) viability. In endothelial nitric oxide synthase (eNOS)-knockout mice, a reduction in nitric oxide bioavailability results in elevated HB-EGF expression, which might be play an important role in progressive diabetic nephropathy. In mice with inactivated HB-EGF gene in the hippocampal area, there is reduced neurogenesis in the subgranular zone (SGZ) of the dentate gyrus. These mice also show suppression of PSD-95 (post synaptic density)/NMDA (N-methyl-D-aspartate)-receptor-NR1 (NMDA receptor)/NR2B subunits, and the subsequent NMDA receptor signaling.

Reconstitution

It is recommended to reconstitute the lyophilized Mouse HB-EGF in sterile 18M-cm H?O not less than 100 μg/mL, which can then be further diluted to other aqueous solutions.
