Biochem/physiol Actions

TH301 is a circadian clock modulator that lengthens a circadian period in cells and tissues. It selectively stabilizes CRY2 through interaction with FAD-binding pocket. TH301 strongly enhanced brown adipocyte differentiation in culture. TH301 showed no or only minor effects on CKIδ, CKIα and CK2 activities in vitro.

CRY1 and CRY2 are essential components of the circadian clock controlling daily physiological rhythms. Accumulating evidences indicate distinct roles of these highly homologous proteins, in addition to redundant functions. Therefore, the development of isoform-selective compounds represents an effective approach towards
