Cell Line Origin

Mouse embryo fibroblast, LRP-deficient, homozygous

Cell Line Description

The cell line PEA-13 is homozygous for a disruption of the gene for low density lipoprotein receptor-related protein (LRP). PEA-13 was derived from fibroblasts from day 15 C57BL/6x129 F1 hybrid mouse embryos. Pseudomonas exotoxin A (PEA) was used to select murine fibroblasts genetically deficient in LRP. Together with the cell lines PEA-10 (Sigma Catalogue number. 97040310) and MEF-1 (Sigma Catalogue number. 98061101) an experimental system is available for the analysis of cellular uptake of functionally diverse ligands and the effect of LRP-deficiency. This is of importance since LRP-deficient mouse embryos die early during gestation.

Application

Study of LRP and LRP deficiency.

Culture Medium

DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).

Subculture Routine

Split sub-confluent cultures (70-80%) 1:6 to 1:10 i.e. seeding 2-4 x 10,000 cells / cm2 using 0.25% trypsin or trypsin/EDTA; 5% CO2; 37°C
