Cell Line Description
The hybrid ND8/34 is a subclone of ND8 which was derived after fusion of primary neonatal rat dorsal-root-ganglion (DRG) neurons with N18Tg2, a mouse neuroblastoma (C1300)-derived azaguanine-resistant line. HAT sensitive hybrids were selected and cloned by limiting dilution. ND8/34 are electrically excitable to the chemical activator of nociceptive sensory neurons, bradykinin.
Application
Study of neuronal differentiation.
Culture Medium
DMEM + 2mM Glutamine + 10% Foetal Bovine Serum (FBS).
Subculture Routine
Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 1-3x10,000 cells/cm2 by either aspirating or gently tapping the cells loose; 5% CO2; 37°C. Cells are lightly attached therefore use of trypsin is unnecessary.
