General description

As a gold standard in hematology, the Harleco® Wright stain is a Romanowsky neutral dye stain based on the combination of methylene blue azure as the basic dye component and eosin as the acidic dye component. The typical purple color of cell nuclei is due to a molecular interaction between eosin Y and an azure B-DNA complex.

This polychromic stain is widely used to identify acidophilic, basophilic, and neutrophilic granules of leukocytes in peripheral blood and bone marrow smears. For decades, the Harleco Wright stain has been used in clinical diagnostics to characterize red blood cell morphology and to differentiate white blood cell types. The staining result can be influenced by several factors such as pH, buffer composition, fixation, and staining time.

The Harleco Wright stain can be used to stain the following sample materials: fresh, native whole blood or bone-marrow smears and clinical cytology specimens (urine sediment, sputum, smears from fine needle aspiration biopsies (FNAB), rinses, and imprints). It may also be used for G-banding to aid in the analysis of poorly spread metaphases.
