General description

FAM163A (family with sequence similarity 163, member A) is a secreted protein, which is also called neuroblastoma-derived secretory protein (NDSP). This gene is located on human chromosome 1q25.2. The encoded protein contains a C-C motif and a signal peptide made of 30 amino acids. It consists of 167 amino acids, and is secreted as both, a monomer and a homodimer. Its molecular weight is 18kDa. It is normally expressed in adrenal gland, brain, thymus, colon, ileum, uterus, and testes, where it has the highest level of expression in testes.

Application

Biochem/physiol Actions

FAM163A (family with sequence similarity 163, member A) is up-regulated in neuroblastoma, as opposed to normal tissues and adult and pediatric cancers. It therefore, plays a role in tumorigenesis of neuroblastoma. This might be achieved by it acting as a ligand for growth factors receptors. It prevents anoikis and supports anchorage independent growth, via ERK (extracellular signal-regulated kinases)-mediated proliferation pathway. FAM163A can therefore, be used as therapeutic target in neuroblastoma. Its mRNA levels also correlate with the risk level in neuroblastoma, and thus, have potential as a marker to determine the metastasis in bone marrow.
