Specificity

Clone 1G19 is a ZooMAb® rabbit recombinant monoclonal antibody that specifically detects Fibrillarin/FBL. It targets an epitope within 15 amino acids from the N-terminal half.

Immunogen

KLH-conjugated linear peptide corresponding to 15 amino acids from the N-terminal half of human Fibrillarin.

Application

Quality Control Testing

Evaluated by Western Blotting in HEK293 cell lysate.

Western Blotting Analysis: A 1:10,000 dilution of this antibody detected Fibrillarin/FBL in HEK293 cell lysate.

Tested Applications

Western Blotting Analysis: A 1:10,000 dilution from a representative lot detected Fibrillarin/FBL in Neuro2a and PC12 cell lysates.

Affinity Binding Assay: A representative lot of this antibody bound Fibrillarin/FBL peptide with a KD of 2.0 x 10-8 in an affinity binding assay.

Immunohistochemistry (Paraffin) Analysis: A 1:100 dilution from a representative lot detected Fibrillarin/FBL in Human small intestine tissue sections.

Immunocytochemistry Analysis: A 1:1,000 dilution from a representative lot detected Fibrillarin/FBL in HEK293 cells.

Note: Actual optimal working dilutions must be determined by end user as specimens, and experimental conditions may vary with the end user.

Target description

rRNA 2′-O-methyltransferase fibrillarin (UniProt: P22087; also known as 34 kDa nucleolar scleroderma antigen, Histone-glutamine methyltransferase, U6 snRNA 2′-O-methyltransferase fibrillarin) is encoded by the FBL (also known as FIB1, FLRN) gene (Gene ID: 2091) in human. Fibrillarin is a highly conserved nucleolar protein that is a component of box C/D small nucleolar ribonucleoprotein (snoRNP) particles that also contain SNU13, NOP5 and NOP56, and a guide RNA. It is one of the most abundant proteins present in nucleoli. It serves as a S-adenosyl-L-methionine-dependent methyltransferase that has the ability to methylate both RNAs and proteins. It is involved in several methylation sites in rRNA that are essential for proper ribosome function. Its site specificity is provided by a guide RNA that base pairs with the substrate. Fibrillarin involved methylation occurs at a characteristic distance from the sequence involved in base pairing with the guide RNA. It is also reported to ACT® as a protein methyltransferase by mediating methylation of Gln-105 of histone H2A (H2AQ104me), a modification that impairs binding of the FACT complex and is specifically present at 35S ribosomal DNA locus. Its acetylation by CREBBP/CBP prevents methylation of Gln-105 of histone H2A, without affecting rRNA methylation. Its deacetylation by SIRT7 restores methylation of H2AQ104me. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, Affinity, reproducibility, and stability over conventional monoclonals. Physical form

Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 µL.

Reconstitution

300 µg/mL after reconstitution at 25 µL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.

Storage and Stability

Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 µL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
