General description

Lectins are carbohydrate-binding proteins. They are omnipresent and usually found in fungi, plants and animals. The secondary structure of this protein is rich in β-strands and possesses carbohydrate binding sites on the surface.

Application

Lectin from Triticum vulgaris (wheat) has been used:
  • to treat transfected cells for immunofluorescence
  • to stereotaxically inject into mice for neuronal tracing
  • in immunohistochemistry for the analysis of myocytes

For the fluorescent detection of glycoproteins containing β(1→4)-N-acetyl-D-glucosamine

Biochem/physiol Actions

Lectin plays a key role in glycoconjugate characterizing, imaging and targeting. Its use in a microarray assay, enable efficient glycome profiling. In plants and fungi, lectin defends against pathogens/feeders. Lectin also plays a major role in host recognition and tissue adhesion.
WGA is not blood group specific but has an affinity for N-acetyl-β-D-glucosaminyl residues and N-acetyl-β-D-glucosamine oligomers. WGA contains no protein-bound carbohydrate.

  1. After reconstitution, how should lectin be stored and for how long at given temperature?

    1 answer
    1. The long term stability of this product in solution has not been investigated. Stock solutions prepared at a minimum of 1 mg/mL in water or PBS may be stored in aliquots up to 3 months at -20 °C when protected from light. Avoid repeated freeze/that cycles.

  2. Please is this product use in staining Arbuscular mycorrhizal fungi in root, for confocal microscopy examination?

    1. This product has not been tested specifically for use in fungal staining in plant tissue. However, FITC-labeled wheat germ agglutinin or lectin has been cited in the literature for such applications. Please see the link below to review one such publication:

  3. Does Sigma provide a stain to visualize cell borders for human Cardiomyocytes derived from the GM23338 iPSC cell line?

    1. There is no recommended stain for cells from this type of cell line. However, for visualizing typical cardiomyocyte cell borders, one may consider L4895, as referenced by Vanhoutte D et al, Nat Commun. 2021 Jun 24;12(1):3928. L4895 is Sigma-Aldrich Lectin from Triticum vulgaris (wheat). The article titled "Thbs1 induces lethal cardiac atrophy through PERK-ATF4 regulated autophagy" in Nat Commun. 2021 Jun 24;12(1):3928 may serve as a point of reference.
