Application

El gel ECM puede utilizarse con células epiteliales, células endoteliales, células musculares, células nerviosas y células tumorales.

Biochem/physiol Actions

El gel ECM experimentará polimerización termoactivada cuando se lleve a una temperatura de 20-40°C, formando una membrana basal reconstituida. Las células PC12 mostrarán formación axónica en dos días si se cultivan en una capa fina de este gel ECM.

Cada colonia de ratones utilizada para la producción de este producto se examina sistemáticamente para detectar varios patógenos. Probado y encontrado negativo para LDEV.

Preparation Note

Este producto se prepara a partir del sarcoma Engelbreth-Holm-Swarm en ratones; dializado contra cloroformo. El gel debe descongelarse durante la noche a 2-8 °C antes de utilizarlo y debe dispensarse a una placa multipared utilizando una placa y pipetas que hayan sido previamente enfriadas a 2-8 °C. El gel también puede diluirse hasta el doble con medio de Eagle modificado de Dulbecco, y debe hacerse antes de añadir el gel a la placa. El producto se gelificará en 5 minutos a 20°C. Para manipulaciones prolongadas, el trabajo debe realizarse por debajo de 10 °C. Las células pueden sembrarse en la parte superior de una fina capa de gel de 0,5 mm o cultivarse en el interior de una capa de 1 mm. Si cultiva las células en el interior, agregue las células al gel antes de colocar en placa a una densidad recomendada de 3-4 x 104 células por ml. Para disociar las células de este gel, utilice proteasa en PBS sin calcio, magnesio o EDTA, a una concentración de 0,6-2,4 unidades por ml.

Other Notes

El ECM se preparó a una concentración proteica de 8-12 mg/ml (en DMEM), que contenía laminina como componente principal, colágeno de tipo IV, proteoglucano sulfato de heparina, entactina y otros componentes menores. La adición de colágeno de tipo IV al gel de ECM aumenta la polimerización.

El producto contiene 50 mg/l de sulfato de gentamicina.

Disclaimer

Para su conservación a largo plazo, mantenga el producto a -20°C. El gel ECM puede conservarse a una temperatura de 2-8°C durante un máximo de 72 horas.

    1. Dispase (Product No. D4818) is a Bacillus-derived neutral protease that is recommended for recovering cells cultured on ECM (Product No. E1270) or Matrigel. Dispase will yield a single cell suspension far more effectively than trypsin, collagenase, or other proteolytic enzymes. Dispase is gentle and will not damage cells harvested for sub-cultivation or other study.
