General description

Upgrading to Wi-Fi connectivity and Millicell® Cloud Access:

Scepter 3.0 instruments purchased before January 1st, 2026, do not have Wi-Fi connectivity. Customers can add Wi-Fi connectivity and get cloud connectivity using the following steps:

1. Download the free software at Millicell® Cloud. Select Scepter 3.0 and download the software onto a USB drive. Load this Software onto the Scepter 3.0 device.

2. Place an order for free trial subscription of Millicell® Cloud using the Catalog number MILLICL-TRIAL. Contact our customer service to help you with this.

3. Purchase Wi-Fi dongle: MERS03USBD0N. Insert the USB dongle into the USB port and follow the instructions on the screen. Now your device is ready to access Millicell® Cloud.

Questions

1–2 of 2 Questions
  1. Will Scepter 3.0 be able to quantify bacterial cultures? How many reads is it possible with a single counter sensor?

    1 answer
    1. No, bacteria are below the size threshold for counting on the Scepter. Sensor tips are designed to be single use.

      Helpful?

  2. Can it count nuclei, specifically isolated from human aortic endothelial cells?

    1 answer
    1. While most mammalian nuclei are around 6 micrometers (um), which is at the low end of detection, this size range can also include other cell debris that is often gated out during analysis. Additionally, since the counts are displayed as a normal distribution, the full profile may not be visible at the low end of detection. Generally, it's not recommended to use this method for small nuclei, but the Scepter may work for some cells with larger nuclei.

    3 out of 5 stars.

Disappointing upgrade from Scepter 2. It has no accompanying analysis software.

As a long term user of the Scepter 2 counter and its accompanying software to monitor the density, size and health of our cells we were pleased to take delivery of a Scepter 3 counter.
While the Scepter 3 counter itself is simple to use and connectivity is good it is the software we use with the Scepter 2 that makes it really useful to us. The Scepter 3 comes with no such software so if you want to compare/overlay histograms you have to extract them from the optional csv files and do it manually. Similarly there is no way to perform a calculation of percentage gated cells (simple measure of viability) without manual extraction and analysis of the data.
This may sound a trivial point but when we are continuously monitoring multiple cell lines at different time points post infection/transfection the software is essential.
Unfortunately the lack of software makes the Scepter 3 no longer an appropriate instrument for our tissue culture analysis requirements.
