General description

Wright Stain is a modified Romanowsky stain commonly used for the cellular elements of blood. Wright′s stain is a mixture of eosin Y with polychromed (oxidized and partly demethylated) methylene blue and therefore termed polychrome stains. The hematological stain is intended for differentially staining peripheral blood and bone marrow films.

Application

Wright stain has been used to determine the number of white blood cell types (neutrophils, lymphocytes, monocytes, eosinophils, and basophils) per microliter of whole blood.
