Description générale

Short chain fatty acids (SCFAs) are saturated fatty acids with less than 6 carbons. SCFAs are produced in the gut by microbial fermentation of dietary fibers.(1) The most important and researched SCFAs are Acetate (Ac), propionate (Pr) and butyrate (Bu).(2) SCFAs are predominantly found in the colon and cross the intestinal epithelium to the blood stream. SCFAs were found to be beneficial and effective in prevention of obesity, Non-alcoholic fatty liver disease and insulin resistance.(3,4)

Application

SCFAs can serve in various applications including in vitro and in vivo administration to study the effect on immune system activation(5), protect epithelial cells from LPS penetration by tight junction formation induction(6), effect insulin secretion(7) and more. In these studies, SCFAs were administrated in vitro at the following concentration range Acetate 1-10mM, propionate and butyrate 0.1-1mM and in vivo Acetate, 70 mM; propionate, 30 mM; butyrate, 20 mM.
Short-chain fatty acids (SCFAs) have diverse roles in various metabolic processes, and their measurement through metabolomics can provide valuable insights into several applications.

This kit contains the following components: 100 mL of 1M Sodium acetate, 100 mL of 0.5 M Sodium propionate, 100 mL of 0.5 M Sodium butyrate, and 1 mL of a short chain fatty acid mix composed of 0.1 M Sodium Acetate, Sodium Propionate, and Sodium Butyrate. Derivation reaction mixtures for detection by LC-MS/HPLC can be prepared as discussed in Table 1 of the technical bulletin.

      Sodium acetate: 100 mL/0.1mL per derivation = 1000 derivations
      Sodium propionate: 100 mL/0.2 mL per derivation = 500 derivations
      Sodium Butyrate: 100 mL/0.2 mL per derivation =500 derivations
      Short Chain Fatty Acid Mix: 1 mL/0.1 mL per derivation = 10 derivations

      Please be aware the procedure provides instructions for HPLC, LC-MS and GC-MS analysis. Sample preparation will vary based upon type of analysis.

    1. The kit components are ready-made aqueous solutions. The concentration of each component included in this kit is as follows:
      1.0 M Sodium Acetate
      0.5 M Sodium Propionate
      0.5 M Sodium Butyrate
      Short Chain Fatty Acid mix – a 0.1 M solution of each above component, Sodium Acetate, Sodium Propionate and Sodium Butyrate, in water.

  3. Respected supplier i would like to know what is the application of the kit? Is this a HPLC standard or is it a kit used for detection of butyrate, acetate and propionate? What will i be receiving in this kit? Can you please be more specific?

    1. This kit may be used for a variety of applications, including calibration of downstream detection of SCFAs by HPLC, GC-MS, or LC-MS. The kit components are ready-made aqueous solutions of 1 M Sodium acetate, 0.5 M Sodium propionate, 0.5 M Sodium butyrate, and the Short chain fatty acid mix.   5. Is this kit free short chain fatty acids? I do not want the sodium attached.

  6. Hello is it possible to use this kit for measuring SCFA in fish? please let me know

    1. Unfortunately, there are no guidelines for using this product to measure short-chain fatty acids in a fish sample. Assuming, there is a means to extract the short-chained fatty acids from a fish sample it may be possible to use this kit. Another concern is that the sample will need to be cleaned up with a sample preparation technique before utilizing LC-MS or GC-MS. It may be possible to use this kit for this purpose, but research and method development would be necessary to determine suitability for the intended application.
