Informations sur le gène

Description générale

Monoclonal Anti-THY1 (mouse IgG1 isotype) is derived from the hybridoma TH350 produced by the fusion of mouse myeloma cells and splenocytes from BALB/c mice immunized with a synthetic peptide corresponding to a sequence at the N-terminus of human THY1, conjugated to keyhole limpet hemocyanin (KLH). Monoclonal Anti-THY1 recognizes human, rat and mouse Thy-1.
Thy-1 cell surface antigen (THY1) is a glycoprotein that is expressed on the membranes of adult cells, mesenchymal stromal cells and cancer cells. It has a molecular weight of about 25-37kDa. The gene encoding THY1 is localized on human chromosome 11q23.3.
THY-1 is expressed in various cell types, including human fibroblasts, neurons, blood stem cells and endothelial cells as well as murine T cells.

Spécificité

Monoclonal Anti-THY1 recognizes human, rat and mouse Thy-1.

Immunogène

synthetic peptide corresponding to a sequence at the N-terminus of human THY1, conjugated to KLH.

Application

Monoclonal Anti-THY1 antibody produced in mouse may be used in:
  • immunoblotting
  • flow cytometry
  • immunofluorescence

Actions biochimiques/physiologiques

THY-1 is also involved in the inhibition of neurite outgrowth, apoptotic signaling, leukocyte and melanoma cell adhesion and migration and fibroblast proliferation and migration. This protein plays a role as a mediator of cell-cell and cell-matrix interactions. Thy-1 is a cell surface marker in identifying local and circulating liver cancer stem cells. This protein′s expression can be regulated by the epigenetic mechanism of promoter region methylation, in which down-regulation of Thy-1 is associated with a more invasive/metastatic clinical phenotype.
Thy-1 cell surface antigen (THY1) has a role in the activation of T-cells, release of neurotransmitters at synapses, enhancement of adhesion of astrocytes, wound healing, fibrogenesis and tumor suppression. It also plays an important role in cell migration and cytoskeletal reorganization.

Forme physique

Solution in 0.01 M phosphate buffered saline, pH 7.4, containing 15 mM sodium azide.

Stockage et stabilité

For extended storage, freeze at -20 0C in working aliquots. Repeated freezing and thawing,or storage in “frost-free” freezers,is not recommended. If slight turbidity occurs upon prolonged storage, clarify the solution by centrifugation before use. Working dilution samples should be discarded if not used within 12 hours.

The precise biological role of Thy-1, a glycophosphatidyl-inositol (GPI)-linked cell surface glycoprotein in non-caveolar lipid raft microdomains, remains enigmatic. Evidence suggests that Thy-1 affects intracellular signaling through src-family protein kinases, and modulates adhesive and migratory events, such as thymocyte adhesion

Thy-1 is a 25-37 kDa glycosylphosphatidylinositol (GPI)-anchored protein involved in T cell activation, neurite outgrowth, apoptosis, tumor suppression, wound healing, and fibrosis. To mediate these diverse effects, Thy-1 participates in multiple signaling cascades. In this review, we discuss Thy-1 signaling
