Application

Le bromhydrate de poly-D-lysine a été utilisé :

  • pour enduire des plaques destinées à la culture de cellules précurseurs gliales de rat (RGPCS).
  • pour enduire des fioles et des boîtes pour la culture de tanycytes.
  • pour préparer la surface de lamelles couvre-objet et faciliter l'adhésion cellulaire.
Les polymères de poly-D-lysine peuvent servir à préparer des surfaces favorisant l'adhérence cellulaire. Les polymères de D-lysine peuvent également être utilisés avec des cellules qui digèrent les polymères de poly-L-lysine et induisent une absorption excessive de L-lysine.

Il est recommandé d'utiliser ce produit comme substrat de culture cellulaire, à raison de 0,5 à 1,0 ml de solution à 0,1 mg/ml pour 25 cm2. Les versions de faible poids moléculaire sont moins visqueuses, mais les versions de poids moléculaire plus élevé offrent davantage de points d'attache par molécule.

Biochem/physiol Actions

Le bromhydrate de poly-D-lysine (PDL) est un facteur d'attachement cellulaire non spécifique qui favorise l'adhésion des cellules aux substrats solides en augmentant les interactions électrostatiques entre les ions chargés négativement de la membrane cellulaire et la surface de culture. Une fois adsorbée sur la surface de culture, la poly-D-lysine augmente le nombre de sites de fixation cellulaire de charge positive.

Preparation Note

Ce produit a un poids moléculaire de 70 000-150 000, est testé sur des cultures cellulaires et est stérilisé par rayonnement gamma. Pour éliminer le HBr, dissoudre le produit dans un tampon neutre et réaliser une dialyse pour retirer les sels. D′une manière générale, pour utiliser ce produit comme facteur de fixation, ajouter 50 ml d′eau stérile de qualité « culture tissulaire » à 5 mg de poly-lysine, et recouvrir en conditions aseptiques la surface avec 1 ml de solution pour 25 cm2. Après 5 minutes, retirer la solution par aspiration et rincer soigneusement la surface. Laisser sécher deux heures avant introduction des cellules et du milieu.

Other Notes

La poly-D-lysine est un polymère d'acide aminé à charge positive comportant environ un HBr par résidu lysine. Le bromhydrate permet de former une poudre cristalline de poly-D-lysine soluble dans l'eau. Une petite quantité de produit peut être repliée en structure bêta, car HBr perturbe la formation de liaisons hydrogène entre les groupements amine et soit les groupements carboxyle soit les fractions contenant des atomes d'azote ou d'oxygène.

Disclaimer

Les solutions stériles sont stables pendant 2 ans maximum lorsqu'elles sont conservées entre 2 °C et 8 °C. À conserver déshydraté à -20 °C.

    1. This product is tested for solubility in water at 50 mg/mL. The product, in solution, is quite stable when stored at 2-8°C. The freeze/thaw stability of this material has not been investigated. However, the thawing process may cause degradation. The product P4832 is a prepared solution that is stored refrigerated and assigned a 2 year shelf life.

  2. Comparitively which is better to use with Human embryonic kidney cells, P6407 or P2636?

    1. Comparison testing of these products on Human embryonic kidney cells has not been conducted. In general, P6407 is preferred due to the fact that P2636, provided in L form, is more susceptible to degradation by proteases secreted by cell lines compared to the d-form polymer provided in P6407.

  3. Is there a standard method of determining the quality of the coating? (Particular focus should be placed on ensuring that the cells adhere as evenly as possible)

    1. As mentioned on the product detail page, this product is recommended as a cell culture substratum when using 0.5 - 1.0 mL of a 0.1 mg/mL solution to coat 25 cm2. Lower molecular weight versions of the product are less viscous, but high molecular weight versions provide more attachment sites per molecule.

  4. Does Product P6407, Poly-D-lysine hydrobromide, need to be dialyzed to remove the hydrogen bromide?

    1. The hydrogen bromide will not interfere with the cell growth.  The hydrogen bromide salts are used to make our poly-L-lysine solutions that are cell culture tested.  For these products, there is no interference in cell growth from the bromide.

  5. How do I determine which molecular weight range of poly-lysine to use?

    1. Poly-lysine is used to coat surfaces to increase the charge present. The lower molecular weight poly-L-lysine (30-70 kD) is easier to use because it is less viscous in solution, but the higher molecular weight poly-L-lysine (>300 kD) provides more attachment sites per molecule. The mid-range product has a molecular weight range of 70-150 kD.  Please check the literature for information on the molecular weight range used for your cell type.

  6. Do you have a protocol for coating cell cultureware or glass coverslips/slides with Product P6407, Poly-D-Lysine?

    1. The protocol for coating surfaces with poly-D-lysine is the same as for poly-L-lysine.  The optimal conditions must be determined for each cell line and application. In general, the following steps can be used.1. Add 50 ml of sterile tissue culture grade water to 5 mg of poly-D-lysine.2. Aseptically coat culture surface with 0.5-1.0 ml of solution per 25 cm2. Rock gently to ensure even coating of the culture surface.3. After 5 minutes, remove solution by aspiration and thoroughly rinse surface with sterile tissue culture grade water.4. Allow to dry at least two hours before introducing cells and medium.

    1. The same protocol can be followed for coating plates with poly-lysine.  The transwell membrane is immersed in the poly-lysine solution.

    1. Certain cells can digest poly-L-lysine, in which case the poly-D-lysine should be used.

  10. How long can I store poly-lysine coated slides or plates?

    1. Plates and/or slides can be stored after drying, protected from dust for up to 1 year.  Inspect the plates/slides to make sure there are no cracks in the coating prior to use.  Each lab should determine the maximum length of time for storage based on the application.
