General description

Un lipopolysaccharide est un glycolipide constitué de glucides liés à des acides gras. Les lipopolysaccharides (LPS) sont des constituants caractéristiques de la paroi cellulaire des bactéries à Gram négatif. Une molécule de LPS typique contient le lipide A (phospholipide de type glucosamine), un oligosaccharide central relativement court et un polysaccharide distal (antigène O). Les LPS contribuent à l'intégrité de la membrane externe et protègent la cellule contre l'action des sels biliaires et des antibiotiques lipophiles. Ils stimulent les cellules hôtes en activant les récepteurs de type Toll (toll like receptors) TLR2 et TLR4. Cette stimulation induit la génération de diverses cytokines pro-inflammatoires, telles que TNFα (tumor necrosis factor, facteur de nécrose tumorale), IL-1 (interleukine 1) et IL-6.
Ce produit est extrait au phénol à partir d'Escherichia coli de sérotype O111:B4. La souche source provient d'une collection privée. Ce sérotype de LPS a été utilisé pour stimuler les lymphocytes B et induire l'oxyde nitrique synthase (NOS, nitric oxide synthase) dans les hépatocytes humains.

Application

Les lipopolysaccharides (LPS) issus de Escherichia coli O111:B4 ont été utilisés :

  • pour stimuler des macrophages.
  • pour un préconditionnement aux LPS chez des souris.
  • pour induire une lésion rénale aiguë chez des souris.
  • pour induire un syndrome de détresse respiratoire aiguë (SDRA) chez des souris afin d'étudier les effets anti-inflammatoires d'exosomes dérivés de cellules souches mésenchymateuses de moelle osseuse (BMSC).
  • pour induire un état hyper-inflammatoire chez des souris infectées par le coronavirus du syndrome respiratoire aigu sévère 2 (SARS-CoV-2).
Les lipopolysaccharides (LPS) sont des éléments constitutifs caractéristiques de la paroi cellulaire des bactéries à Gram négatif. Les LPS et leur fraction lipidique (lipide A) stimulent les cellules du système immunitaire inné par l'intermédiaire du récepteur TLR4 (Toll-Like Receptor 4), qui appartient à la famille des récepteurs de type Toll et qui reconnaît la signature moléculaire des principaux agents pathogènes (PAMP).

Biochem/physiol Actions

Les lipopolysaccharides (LPS) constituent une partie de la membrane externe, ce qui rend cette dernière perméable aux molécules hydrophobes telles que les substances antimicrobiennes qui procurent une résistance innée aux bactéries à Gram négatif. Les LPS agissent sur les interactions bactérie-hôte qui régulent les réponses du système hôte.

Preparation Note

Ce produit est soluble dans l′eau (5 mg/ml) ou dans du milieu de culture cellulaire (1 mg/ml) et donne une solution trouble jaune pâle. Une solution plus concentrée (20 mg/ml), toujours trouble, a été obtenue dans une solution saline aqueuse après mélange au vortex et chauffage à 70-80 °C. Les lipopolysaccharides sont des molécules qui forment des micelles dans tous les solvants. Des solutions troubles sont observées dans l′eau et la solution tampon phosphate. Les solvants organiques ne produisent pas une solution plus limpide. Le méthanol donne une suspension turbide avec des particules flottantes, tandis que l′eau produit une solution trouble homogène.

  2. Can I resuspend this LPS in PBS instead of water, without hindering its performance ?

    1. This product can be resuspended in PBS instead of water. Both water and PBS are suitable solvents, and a hazy solution will form in either.

  3. How can I determine the shelf life / expiration / retest date of this product?

    1. If this product has an expiration or retest date, it will be shown on the Certificate of Analysis (COA, CofA). If there is no retest or expiration date listed on the product's COA, we do not have suitable stability data to determine a shelf life. For these products, the only date on the COA will be the release date; a retest, expiration, or use-by-date will not be displayed.
      For all products, we recommend handling per defined conditions as printed in our product literature and website product descriptions. We recommend that products should be routinely inspected by customers to ensure they perform as expected.

  4. When I compared COAs of two batches of LPS (L2630) we have been using, I realized the potency of the newer one is 10-fold higher than the older one (30000000 and 3000000 EU/mg). Is it common for LPS that different lots have such different potency?

    1. Significant lot-to-lot variability in potency is quite normal for this product. It is a somewhat crude phenol extract; it is not chromatographically purified. That is, a chromatographically purified LPS such as Product L3012 would be expected to have better lot-to-lot consistency. Note that for both this product and L3012, the potency specification is greater than or equal to 500,000 Endotoxin units per milligram (EU/mg); there is a lower limit, but there is no upper limit.

  5. How is shipping temperature determined? And how is it related to the product storage temperature?

    1. Products may be shipped at a different temperature than the recommended long-term storage temperature. If the product quality is sensitive to short-term exposure to conditions other than the recommended long-term storage, it will be shipped on wet or dry-ice. If the product quality is NOT affected by short-term exposure to conditions other than the recommended long-term storage, it will be shipped at ambient temperature. As shipping routes are configured for minimum transit times, shipping at ambient temperature helps control shipping costs for our customers.   6. Is there any harm in filter sterilizing a solution with this LPS before using it in cell culture or animals? Meaning does it change or lose potency if it is sterile filtered say through a 0.2 micron filter?

Sterile filtration should not affect the concentration or properties of the solution if the LPS is fully dissolved, but please keep in mind that this is a research-grade material that has not been best tested on animals.   7. I received LPS L2630 at Room Temperature. Is it ok to keep it at Room Temperature before reconstitution, although recommendations are to store it between 2-8 C?

    1. This product ships routinely at ambient temperature and is stable up to 2 weeks to allow for transit time. The item is to be stored at 2-8°C when received.
