Description

General description

Recombinant human Hepatocyte Growth Factor (HGF) is supplied as a lyophilized protein suitable for mammalian cell culture research. HGF is a multifunctional growth factor that signals through the c‑Met receptor and is widely used in in vitro cell studies.

Application

HGF has been used in mammalian cell culture research applications, including epithelial, endothelial, and hepatocyte‑related in vitro model systems.

Biochem/physiol Actions

Hepatocyte growth factor (HGF) is a potent mitogen for hepatocytes and a variety of cell types including endothelial and epithelial cells, melanocytes, and keratinocytes. It also mediates epithelial morphogenesis. HGF is the ligand for the tyrosine kinase receptor encoded by the met proto-oncogene.

Hepatocyte Growth Factor (HGF) exhibits mitogenic, motogenic, and morphogenic activity in vitro across multiple cell types through activation of c‑Met receptor‑mediated signaling pathways.

Physical form

Lyophilized from a 0.2 μm filtered solution in 20 mM NaH₂PO₄ and 500 mM NaCl, pH 7.0 with 50 μg BSA per 1 μg HGF

Preparation Note

Reconstitute at 50 μg/mL in sterile PBS containing at least 0.1% human or bovine serum albumin. After reconstitution, this solution can be stored at 2-8 °C for up to one month or at -20 °C to -70 °C for up to three months. Us a manual defrost freezer and avoid repeated freeze-thaw cycles.

Analysis Note

The biological activity is measured by its ability to induce IL-11 secretion by Saos-2 human osieosarcoma cells.

Other Notes

The NSO-expressed recombinant HGF is greater than 95% heterodimeric HGF. The insect cell line-expressed recombinant HGF is a mixture of single chain HGF (predominant) with some heterodimeric HGF. Single chain HGF and heterodimeric HGF are equally active in in vitro assays.
