The cell line OE33, also known as JROECL33, was established from the adenocarcinoma of the lower oesophagus (Barrett′s metaplasia) of a 73 year old female patient. The tumour was identified as pathological stage IIA (UICC) and showed poor differentiation. OE33 express HLA-A, -B and -C antigens (MHC class I) and ICAM-1 constitutively. Expression of HLA-DR (MHC class II) can be induced by treatment with interferon-gamma. The cells express epithelial cytokeratins and are tumourigenic in nude mice.

Application

OE33 cell line has been used to study the effect of microRNA-MiR-193b on autophagy and cell death on these cells.[1] It has also been used to study the sensitivity of the cells to anti-c-Met monoclonal antibody ABT-700.[2]

RPMI 1640 + 2mM Glutamine + 10% Foetal Bovine Serum FBS / FCS.

Procédure de repiquage

Split sub-confluent cultures (70-80%) 1:8 i.e. seeding at 1x10,000 cells/cm2 using 0.25% Trypsin or Trypsin/EDTA; 5% CO2; 37°C. Initially these cells grow slowly and can take up to 7 days until ready for the next split, 50% media changes will be necessary every 2-3 days (i.e. replacing half the old medium with fresh).

  1. How do I handle Cell Line frozen cells upon arrival?

    1 answer
    1. Upon receipt, frozen ampules should be transferred directly to vapor phase liquid nitrogen without delay (-135°C) or liquid phase liquid nitrogen if vapor phase is not available. DO NOT use a -80°C freezer as an alternative; this will result in loss of viability.

  2. How many Cell Line cells are in the vial?

    1 answer
    1. Each vial of cells contains 2-3 x 106 cells in 1 ml of freezing media. This is in a 1.8 mL ampule.  For exceptionally large cells, counts may decrease. Suspension cells, generally smaller cells, may contain as many as 4-5 x 106 cells/vial to assure optimal viability upon thaw.

  3. What medium should I use when I thaw the Cell Line cells?

    1. The medium for each cell line is listed on the product page. It is specific to each cell line. If not visible on the Sigma-Aldrich website product page, the HPA website also contains the same information (www.hpacultures.org.uk). The product number for the cell line is the same on either site.

  4. Why is Vapor Phase Liquid Nitrogen preferred for storage of Cell Line cells?

    1. If ampules are immersed into liquid phase of liquid nitrogen, it increases the risk of the liquid seeping into the vial. This could lead to problems of cross-contamination and increased risk of the ampule exploding when thawed.

    1. ECACC routinely tests all manufactured cell banks for mycoplasma.
