Description générale
stabilisée au tartrate double de potassium et de sodium). L′ajout du réactif phénolique génère des chromogènes qui augmentent l′absorbance entre 550 et 750 nm. Typiquement, le pic d′absorbance (750 nm) ou son épaule (660 nm) sont utilisés pour déterminer la concentration des protéines entre 1-100 mg/ml, tandis que l′absorbance à 550 nm est utilisée pour déterminer les concentrations plus élevées.
En l′absence de cuivre, l′intensité de la coloration est déterminée principalement par le contenu en tyrosine et en tryptophane des protéines, et dans une moindre mesure en cystéine et en histidine. Le cuivre(II) n′a aucun effet sur la formation d′une coloration par la tyrosine, le tryptophane ou l′histidine, mais réduit la formation de coloration par la cystéine.[1][4][5][6][7]
De nombreuses modifications de la procédure initiale du dosage ont été publiées[4], dont des méthodes pour accélérer le développement de la coloration[6][8], pour déterminer la teneur en protéines insolubles[1][9] et pour automatiser la procédure.[10] Plusieurs substances comme de nombreux tampons, agents chélateurs, détergents et composés organiques cycliques peuvent interférer avec le dosage des protéines selon Lowry.[4]
Le réactif phénolique de Folin-Ciocalteu peut également être utilisé comme réactif en aérosol dans les procédures chromatographiques.

How do I differentiate the spectrophotometric result obtained from this reagent is either because of reaction with phenols or non-phenolic reducing substances? Thanks.

The type and amount of non-phenolic compounds present is a given sample will determine the impact on the total phenol values. The use of a standard curve is critical. Factors such as pH adjustment can minimize the effects. Reducing compounds may be identified and quantified separately or removed from the sample by various means.
