General description

Fluorescein is a xanthene dye and is a common labeling agent in biology.

Application

Fluorescein sodium salt has been used as a solute to study microvessel permeability. It has been used as a test molecule to study blood barrier penetration.
Water soluble fluorescent dye used in permeability and vascular perfusion studies due to its small molecular radius.[1] Sodium fluorescein is used as a fluorescent tracer to study blood-brain barrier (BBB) and blood-spinal cord barrier (BSCB) permeability in rodent models.[2][3][4] The dye was used as a probe substrate in the study of organic anion transporting polypeptide (OATP) mediated drug transport in hepatocytes.[5] Sodium fluorescein was also used to monitor in situ solute transportation into bone through fluorescence recovery after photobleaching (FRAP).[6]

  1. Can I make a stock solution of this in HBSS? If yes, what is the stability and recommended storage conditions?

    1. For product F6377 the dye is tested in water at a low concentration. Conn's Biological Stains, 10th Edition states the sodium salt in soluble in water up to 50%. It is expected the dye should dissolve in HBSS but there is no supportive data. There is also no solution stability information available. Although the dry powder should be stored at room temperature, solutions are probably best kept at 2-8°C.

  2. Hello, I would like to ask if this product's fluorescence can be quenched by paraformaldehyde (PFA)? Thank you.

    1. The preferred fixative for various staining procedures is often freshly prepared paraformaldehyde solution. Paraformaldehyde solution is not known to quench fluorescein sodium salt or other fluorescent molecules. However, no specific analysis been performed as part of the quality control testing performed on product F6377 to ensure fluorescence is not quenched.

  4. Can this product be autoclaved once dissolved? How long can a concentrated dissolved stock solution be stored for?

    1. The stability of Fluorescein under autoclave conditions has not been determined. Solutions have typically been filter sterilized. Long term solution stability has also not been evaluated. However, solutions of Fluorescein Diacetate are stable at -20°C in ethanol when protected from light. Similar stability would be expected with the sodium salt form.

    1. Unfortunately, a pharmacological grade Fluorescein or Fluorescein sodium salt is not available at this time.

  6. Is F6377 soluble in ethanol?

    1. The substance is soluble in water (500 g/l) at 20°C and slightly soluble in ethanol, but insoluble in benzene, chloroform, and ether.
