General description

POU class 5 homeobox 1 (POU5F1) recognizes defined octamer DNA element by a shared, conserved DNA-binding domain, namely the POU domain.
POU class 5 homeobox 1 (POU5F1), also known as octamer-binding transcription factor-4 (OCT-4), is expressed in embryonic stem cells and germ cells. It belongs to the POU family of transcription factors. Three splice variants of the protein (OCT4A, OCT4B and OCT4B1) have been identified. The gene encoding this transcription factor is localized on human chromosome 6p21.33.

Immunogen

synthetic peptide corresponding to amino acids 19-34 of human POU5F1, conjugated to KLH. The corresponding sequence in rat is 81% homologous.

Application

Anti-POU5F1 (Oct4) antibody produced in rabbit has been used in Western Blotting.

Biochem/physiol Actions

POU class 5 homeobox 1 (POU5F1) may be a tumor initiating stem-like cell (TSC)-related marker. It has a role in stem cell pluripotency and self-renewal, and also modulates differentiation. The protein has been shown to be expressed in several cancers. POU5F1 is also vital for the induced pluripotent stem cell (iPSC) process.
POU5F1 or Oct4/Oct3 (Octamer-binding transcription factor), functions as an early developmental control gene. It has been recognized as fundamental in the maintenance of pluripotency in embryonic stem cells and in primordial germ cells. It is down regulated in all differentiated somatic cell types both in vivo and in vitro. This is being accomplished through increased DNA methylation and structural changes involving the immediate upstream regulatory region. POU5F1 serves as a marker for germ cell tumors (GCTs) eliciting pluripotentiality, especially including embryonal, seminoma/ dysgerminoma/ germinoma.

Solution in 0.01 M phos­phate buffered saline, pH 7.4, containing 15 mM sodium azide.
