General description

PRKC apoptosis WT1 regulator protein (PAWR) is a 32-amino acid protein with a leucine zipper domain at its carboxy terminus. The gene encoding it is located on chromosome 12q21. PAWR is commonly referred as PAR4 (Prostate apoptosis response 4 protein).

Application

Biochem/physiol Actions

PRKC apoptosis WT1 regulator protein (PAWR) induces the apoptosis of prostate cancer cells and causes tumor regression. It inhibits the nuclear factor κ-B (NF-κB) activity and also the cell membrane trafficking of Fas (a cell surface death receptor) and Fas ligand (FasL). This leads to the activation of the Fas-Fas-associated death domain-caspase-8 pro-death pathway. The deletion in the gene encoding PAWR leads to pancreatic cancers and male germ cell tumors. PAWR takes part in neuronal degeneration and cholinergic signaling in Alzheimer′s disease. It interacts with CHT1 (a choline transporter) and regulates choline uptake. It functions as a negative regulator of the choline uptake activity by reducing the CHT1 incorporation on the cell surface. WT1 (a transcriptional regulator) has a transcriptional activation domain which can only be activated by specifically interacting with PAWR.
