General description

Tyrosine 3-monooxygenase (EC 1.14.16.2; UniProt P04177; also known as TH, Tyrosine 3-hydroxylase) is encoded by the Th gene (Gene ID 25085) in rat species. Tyrosine hydroxylase plays an important role in the physiology of adrenergic neurons. It is the first and rate-limiting enzyme involved in the biosynthesis of the catecholamines, dopamine and norepinephrine, from tyrosine. TH is, therefore, a useful marker for dopaminergic and noradrenergic neurons. TH enzymatic activity requires ferrous ions as cofactor and is believed to be regulated by phosphorylation. Six human TH isoforms exist as a result of alternative splicing (UniProt P07101-1 through P07101-6).

Specificity

Clone 10F8.2 targets a C-terminal epitope present in rat and mouse TH sequence (UniProt P04177 & P24529), as well as in all six human TH spliced isoforms (UniProt P07101).

Epitope: C-terminal region.
KLH-conjugated linear peptide corresponding to a C-terminal region sequence of rat tyrosine hydroxylase.

Application

Immunohistochemistry Analysis: A 1:250 dilution from a representative lot detected tyrosine hydroxylase in rat brain tissue sections.

This Anti-Tyrosine Hydroxylase Antibody, clone 10F8.2 is validated for use in Western Blotting, Immunohistochemistry (Paraffin) for the detection of Tyrosine Hydroxylase.

Quality

Evaluated by Western Blotting in PC12 cell lysate.

Western Blotting Analysis: 0.25 µg/mL of this antibody detected tyrosine hydroxylase in 10 µg of PC12 cell lysate.

Target description

~50-60 kDa observed. 55.83 kDa (rat), 55.86 kDa (mouse), 58.03/55.48/58.47/55.92/45.21/44.77 kDa (human isoform 1/2/3/4/5/6) calculated (Met1 removed). The multiplet banding pattern observed in rat PC12 lysate most likely represent differentially phosphorylated TH species (Gozal, E., et al. (2005). J. Appl. Physiol. 99(2):642-649).
