General description

Native trypsin from bovine pancreas. Serine protease that specifically hydrolyzes amides, peptides and proteins at the C-terminal side of arginine and lysine residues. Inhibitors include aprotinin, DFP, and soybean trypsin inhibitor. Note: 1 MU = 1,000,000 units.

Note: 1 NF unit = 1 USP unit = 3 SU at 30°C, pH 7.6; 1 TAME unit = 19.2 USP units = 57.5 BAEE units; 1 MU = 1,000,000 units.

Unit Definition

One USP unit is defined as the amount of enzyme that will digest BAEE and cause an increase in absorbance of 0.003 units at 253 nm per min at 25°C, pH 7.6.

Reconstitution

Following reconstitution in HCl, aliquot and freeze (-20°C) for long-term storage or refrigerate (4°C) for short-term storage. Dilute into aqueous buffers (at neutral pH) just prior to use. Following reconstitution in H₂O or buffer, use immediately or aliquot and freeze (-70°C) to minimize autolysis. HCl stock solutions are stable for up to 1 week at 4°C or for up to 1 month at -20°C. H₂O or buffer stock solutions are stable for up to 1 month at -70°C.
