General description

Irreversible inhibitor of serine proteases. Its mechanism of action is analogous to that of diisopropylfluorophosphate. PMSF causes sulfonylation of the active-site serine residues. Also reported to inhibit internucleosomal DNA fragmentation in immature thymocytes. For a related, more stable inhibitor, see AEBSF (Cat. No. 101500).

Preparation Note

Following reconstitution, refrigerate (4°C). Stock solutions are stable for up to 9 months at 4°C.

  1. What should it be reconstituted in for best results? You mention the short half life yet long stability when refrigerated.

    1. Phenylmethylsulfonyl Fluoride can be dissolved in either anhydrous isopropanol, ethanol or DMSO at either 100 mM or 200 mM. Aliquot in single use aliquots and store at-20°C for 6 to 9 months. Protect from moisture. The recommended storage temperature and length of storage is from literature references and not from data collected by Millipore Sigma.

      Once the concentrated stock is diluted, prepare in a suitable aqueous buffer and use immediately. Do not refrigerate the working solution for future use.
