Target description
Collagen alpha-1(X) chain (UniProt: Q03692) is encoded by the COL10A1 gene (Gene ID: 1300) in human. Collagen is the most abundant protein in the body that is predominantly synthesized by fibroblasts. However, epithelial cells may also synthesize small amounts of collagen. The collagen super family includes over 20 different types of collagen with at least 38 distinct polypeptide chains. Collagen X a1 is a homotrimeric protein that is synthesized with a signal peptide (aa 1-18), which is subsequently cleaved off to produce the mature protein that contains two non-helical regions (aa 19-56 and 520-680) and one triple helical region (aa 57-519). Type X a1 collagen is a product of hypertrophic chondrocytes and is localized to mineralization zones of hyaline cartilage. It is a major component of hypertrophic zone and it also influences deposition of other matrix molecules to this region. This provides a proper environment for hematopoiesis, mineralization, and modeling that are essential for endochondral ossification. It is considered as a specific marker of hypertrophic chondrocytes and altered expression of COL10A1 gene is often accompanied by abnormal chondrocyte hypertrophy in many skeletal diseases. Mutations in this gene are reported in Schmid type metaphyseal chondrodysplasia (SMCD), an inherited disorder of the osseous skeleton with short stature, sclerosis of the ribs, and flaring of the metaphyses. Significantly higher expression of COL10A1 gene has also been reported in colorectal cancers. This ZooMAb® recombinant monoclonal antibody, generated by our propriety technology, offers significantly enhanced specificity, affinity, reproducibility, and stability over conventional monoclonals. Physical form
Purified recombinant rabbit monoclonal antibody IgG, lyophilized in PBS with 5% Trehalose, normal appearance a coarse or translucent resin. The PBS/trehalose components in the ZooMAb® formulation can have the appearance of a semi-solid (bead like gel) after lyophilization. This is a normal phenomenon. Please follow the recommended reconstitution procedure in the data sheet to dissolve the semi-solid, bead-like, gel-appearing material. The resulting antibody solution is completely stable and functional as proven by full functional testing. Contains no biocide or preservatives, such as azide, or any animal by-products. Larger pack sizes provided as multiples of 25 μL.
Reconstitution
300 μg/mL after reconstitution at 25 μL per vial. Please refer to guidance on suggested starting dilutions and/or titers per application and sample type.
Storage and Stability
Recommend storage of lyophilized product at 2-8°C; Before reconstitution, micro-centrifuge vials briefly to spin down material to bottom of the vial; Reconstitute each vial by adding 25 μL of filtered lab grade water or PBS; Reconstituted antibodies can be stored at 2-8°C, or -20°C for long term storage. Avoid repeated freeze-thaws.
