Unconventional prefoldin RPB5 interactor (URI) is an oncogene required for the survival of some human carcinomas and is often amplified and over-expressed in subsets of carcinomas from ovary, breast, stomach, and lung. Activation of URI contributes an important mechanism for activating mitochondrial S6K1-BAD signaling and promoting cell survival through disabling PP1γ-dependent negative feedback inhibition. URI can negatively modulate RNA polymerase II subunit 5 (RPB5) and mediates resistance of cells to cisplatin.
